Ruiko Sakuragi (桜樹 るい子 (さくらぎ るいこ) Sakuragi Ruiko) is a high school student. She is the former president of the Mystery Club before Makoto Makabe and Miyuki Nanase.

She has a sensual atmosphere, and she sometimes mutters suggestive erotic lines. She has her breasts emphasized in the anime version. She said in her own words that she likes man who's handsome and intelligent.
She is good at seeing through people's qualities, and is one of the few people who has been aware of Kindaichi's ability (in that respect, it is the same as Ogata-sensei in the manga version of Opera House Murder Case).

- The name Ruiko means "child" (子) (ko).
- Ruiko's surname Sakuragi means "cherry blossom" (桜) (sakura) and "tree" (樹) (ki/gi).
- Part of the surname Sakura can also mean "blossom" (咲) (saku) and "good, virtuous, respectable" (良) (ra).
